Video footage obtained by Channel Nine showed detectives driving a black Mercedes-Benz from her Avondale Heights home towards the Calder Freeway, before moving onto the Melton Highway on Thursday.
It is believed police were following a route the vehicle took between her home and the Watergardens shopping centre, where she owned a business with her husband, Borce.
The report follows the towing of a black Mercedes-Benz from the 47-year-old's house on Monday.
Neighbours saw the car, sealed with yellow tape, being loaded onto a tow truck outside Ms Ristevski's home at 1.20pm.
An unmarked police car with flashing lights escorted the truck away from the home.
Ms Ristevski went missing on June 29 last year.
Her body was found between two logs in the Macedon Regional Park on February 20, seven months later.
A new search of Mount Macedon was conducted earlier this month after police received fresh information about the case.
Victoria Police spokesman Chris Metevelis confirmed Thursday that detectives were testing a black Mercedes in the Avondale Heights and Watergardens areas.
However, he said that it would be "inappropriate to comment further", since this was an ongoing investigation.
